The story starts off at the end of DP, which was refreshing. The gen 3 aesthetic took a moment to get used to at first (since we're used to seeing Diamond and Pearl in, well, a gen 4 style), but it fit surprisingly well and executed brilliantly. Pokémon Diamond and Pearl being the first games I ever played (and which I played a lot), I unconsciously went into this game with a high standard - and I was not disappointed. Just to drive it home, I really enjoyed this game, and I recommend to anyone who enjoyed Diamond and Pearl to play it. It is a tiny thing and I imagine part of it comes down to the game jam restraints. Felt a bit weird having pools of water in the game, being the champion, having access to the move surf, being able to use strength and fly, but not being able to use surf. It just felt a bit weird seeing a NPC complaining about the intense weather when it looked normal. Before you take down the roaming pokemon, there are no weather effects on the maps, but the npcs still talk about them. But being able to chuck a ground type against thunderous or a flying type against landerous makes the fight a lot easier when they keep using moves which don't do anything just by luck. The AI of the bosses could be a bit funky at time, I guess since it uses the wild pokemon AI. These aren't really complaints as much as they are things that just felt a bit weird? So yeah, don't take them too harshly. Really makes the battle harbour complete. And a nice touch of polish to the game is just how many competitive items are included hidden around the map. Honestly it isn't really my thing, I've never been too into the battle facilities, but I really appreciate that a fan game made one. Normally a legendary battle in Pokemon games can be pretty easy and luck luster, but the way these boss battles were done made it feel so cool. The actual boss battles were amazing, really drives home the boss battle vibe. First of all, all the puzzles were fun, new and inventive. The dungeons/boss battles are really where this game peaks. Alto town was so cool, I really enjoyed arriving at this town and being whaaa?!?! The second thing I bear in mind is that even if you have highly sensitive system, you should still make sure that what you compare have been done in one experiment and analyzed in one-shot to decrease the possibility of error or bias. So I would doubt the amount inference made by ImageJ intensity analysis, let alone performing statistical analysis afterwards under our lab setting. The trends were right, definitely greater intensity with more loading, but it was not two-folds of intensity in 4ug, or 4 folds in 8ug compared to 2ug loading. Then he did the blot and analyzed with ImageJ and he didn't see the intensity increase the same folds as he loaded in the first place. Actually one member of our lab once tried the loading control, where he loaded the same protein lysates at scaler amounts, say, 2ug, 4ug, 8ug,20ug. There are multiple choices outside and what we use in our lab is traditional film system and it's not so sensitive to distinguish slight difference. We recently had to perform a couple (always trying to please grant reviewers) and while I was analyzing the data using ImajeJ I started to wonder the exact same thing. I find this post extremely interesting, my lab is a physiology lab and we try not to do western blots unless we absolutely have to. Since your analysis says there is, it is important to know if this difference is biologically relevant. Again, this is all just me observing this band without knowing the scientific relevance (or really the experiment at all), but when I look at those bands I do not see a difference in intensities. This also brings to light a point that sometimes we rely to heavily on the "statistical significance" of these band intensities. Or if it would just be another layer of area selection bias. Me wonder if the loading control would help. Regardless your analysis could shows the consequences of these types of analyses, and it makes ![]() I realize this was just a quick experiment of intrigue, and it looks like maybe that top band is the control, or a spurious band. I am curious if you did the same analysis with a loading control, and then normalized the signal. Something like a technical replicate of the area selection? As you mentioned, it is important to correct for noise and that is another added step of variance and potential bias. It makes sense that there is variation between your attempts because of the area around the band, and I wonder if it would be possible to take that into consideration when you are doing western blot analysis. This is a really interesting analysis, that I have always wondered about. |
